Abstract: This Podcast features a conversation with the senior author of a Research Article published in the 11 October 2011 issue of Science Signaling. Eddy De Robertis discusses his groups finding that the phosphorylation status of the transcription factor Mad determines whether it participates in Wingless (Wg) or bone morphogenetic protein (BMP) signaling (1). Phosphorylation of Mad by the BMP receptor leads to Mad-mediated stimulation of BMP-responsive target genes, whereas phosphorylation of Mad by glycogen synthase kinase 3 (GSK-3) targets it for degradation. Wg signaling prevents GSK-3–mediated phosphorylation of Mad, thus allowing it to stimulate expression of Wg-responsive genes.
