Abstract:
Mitochondrial outer membrane permeabilization by proapoptoticBcl-2 family proteins, such as Bax, plays a crucial role inapoptosis induction. However, whether this only causes the intracytosolicrelease of inducers of caspase-dependent death, such as cytochromec, or also of caspase-independent death, such as apoptosis-inducingfactor (AIF) remains unknown. Here, we show that on isolatedmitochondria, Bax causes the release of cytochrome c, but notof AIF, and the association of AIF with the mitochondrial innermembrane provides a simple explanation for its lack of releaseupon Bax-mediated outer membrane permeabilization. In cellsoverexpressing Bax or treated either with the Bax- or Bak-dependentproapoptotic drugs staurosporine or actinomycin D, or with hydrogenperoxide, caspase inhibitors did not affect the intracytosolictranslocation of cytochrome c, but prevented that of AIF. Theseresults provide a paradigm for mitochondria-dependent deathpathways in which AIF cannot substitute for caspase executionersbecause its intracytosolic release occurs downstream of thatof cytochrome c.
