Regeneration of Male Germline Stem Cells by Spermatogonial Dedifferentiation in Vivo

Abstract:
Although the ability of engrafted stem cells to regenerate tissuehas received much attention, the molecular mechanisms controllingregeneration are poorly understood. In the Drosophila male germline,local activation of the Janus kinasesignal transducerand activator of transcription (Jak-STAT) pathway maintainsstem cells; germline stem cells lacking Jak-STAT signaling differentiateinto spermatogonia without self-renewal. By conditionally manipulatingJak-STAT signaling, we find that spermatogonia that have initiateddifferentiation and are undergoing limited mitotic (transit-amplifying)divisions can repopulate the niche and revert to stem cell identity.Thus, in the appropriate microenvironment, transit-amplifyingcells dedifferentiate, becoming functional stem cells duringtissue regeneration.
