Modulation of Hematopoietic Stem Cell Homing and Engraftment by CD26

Abstract:
Hematopoietic stem cell homing and engraftment are crucial totransplantation efficiency, and clinical engraftment is severelycompromised when donor-cell numbers are limiting. The peptidaseCD26 (DPPIV/dipeptidylpeptidase IV) removes dipeptides fromthe amino terminus of proteins. We present evidence that endogenousCD26 expression on donor cells negatively regulates homing andengraftment. By inhibition or deletion of CD26, it was possibleto increase greatly the efficiency of transplantation. Theseresults suggest that hematopoietic stem cell engraftment isnot absolute, as previously suggested, and indicate that improvementof bone marrow transplant efficiency may be possible in theclinic.
