Abstract:
Lysosomes are organelles central to degradation and recyclingprocesses in animal cells. Whether lysosomal activity is coordinatedto respond to cellular needs remains unclear. We found thatmost lysosomal genes exhibit coordinated transcriptional behaviorand are regulated by the transcription factor EB (TFEB). Underaberrant lysosomal storage conditions, TFEB translocated fromthe cytoplasm to the nucleus, resulting in the activation ofits target genes. TFEB overexpression in cultured cells inducedlysosomal biogenesis and increased the degradation of complexmolecules, such as glycosaminoglycans and the pathogenic proteinthat causes Huntingtons disease. Thus, a genetic programcontrols lysosomal biogenesis and function, providing a potentialtherapeutic target to enhance cellular clearing in lysosomalstorage disorders and neurodegenerative diseases.
