Physiology SOCS is the Heavy

Some cytokine actions can be thwarted by members of the suppressors of cytokine signaling (SOCS) family. However, when mice lacking SOCS-2 were generated, they did not exhibit any hematological abnormalities, but rather, grew larger in overall size compared with their wild-type littermates; the SOCS2 null adults were 40% heavier than the wild-type. The mass of several organs and body length increased because of an increase in cell number rather than cell size. This phenotype closely resembled that of mice with increased insulin-like growth factor I (IGF-I) production. Metcalf et al. found that the SOCS-2 null mice had increased IGF-I production in several of the enlarged organs and propose that SOCS-2 may be a negative regulator of IGF-I signaling. This supports previous work in cultured cells that reported the inhibitory effect of SOCS-2 on growth hormone signaling.
