Immunology Adapting the Adaptor

Adaptor proteins connect the proteins involved in multiple cell signaling pathways. In T cells, the adaptor protein ADAP regulates certain signals from the T cell receptor (TCR) that influence the function of integrin adhesion receptors. Medeiros et al. found that ADAP also associates directly with another adaptor, CARMA1, a membrane-associated protein that couples TCR signaling to the activation of the core transcription factor NF-{kappa}B. This association involved the assembly of a multiprotein complex that failed to form in the absence of ADAP and corresponded with impaired NF-{kappa}B activity. The study introduces a key new step in regulating the pathway that connects TCR activation with the transcriptional response to infection.
