A model with Muncs. Syntaxin, freed by NSF from nonproductive complexes (as shown) binds in a closed conformation to Munc18. Munc13 chaperones the assembly of ternary v/t-SNARE complexes that contain syntaxin, SNAP-25, and synaptobrevin. Synaptotagmin-Ca2+ is required for efficient membrane fusion.
