Phosphorylation-Mediated Neurosteroid Actions

Neurosteroids are produced in the brain and regulate the activity of neurons. Allopregnanolone (ALLO) is a neurosteroid that facilitates GABAA receptor-mediated currents. Fáncsik et al. found that ALLO increased the decay phase of spontaneous GABA currents in rat hypothalamic slice preparations. The ALLO-induced increase in the decay phase was blocked by pharmacological treatments that inhibited the activity of protein kinase C (PKC) or GTP-binding proteins (such as those downstream of the G protein-coupled receptors). The authors suggest that in magnocellular neurons the neurosteroid effect requires phosphorylation of the GABAA receptor.
