Looks and Acts Like Sprouty

Two proteins identified by Wakioka et al. have been named Spred-1 and Spred-2 because they contain a Sprouty domain (SPR) and an Ena/VASP homology domain. Like Sprouty, the Spred proteins appear to inhibit growth factor-mediated stimulation of the Ras-MAP kinase pathway, and when overexpressed, they block differentiation of a neuron and muscle cell line. Endogenous Spred and Ras were found in a complex and also colocalized at the cell surface. Overexpression of Spred increased Ras-Raf interaction. The authors postulate that a Spred-induced increase in Ras-Raf interaction may somehow inhibit Raf activation.
