Reassessing Hippocampal Mossy Fiber LTP

It has become generally agreed that the expression of long-term potentiation (LTP) at mossy fiber-CA3 synapses in the hippocampus is a presynaptic phenomenon. The LTP induction site, however, is still controversial. Contractor et al. used intracellular infusion of peptides and antibodies, as well as extracellular ligand application, to demonstrate a role of postsynaptic interaction of GRIP protein with ephrin B receptors in the induction of LTP. Their data suggest that a retrograde signaling pathway, an ephrin B receptor-B ephrin ligand interaction, links postsynaptic induction with presynaptic expression of long-term changes in neurotransmitter release probability.
