A key question in cell and developmental biology is, how do signaling pathways regulate cell function? New tools enable the observation and manipulation of cellular signaling in real time. Studies using these techniques hold the promise of revealing the role of dynamics in controlling the output of signaling pathways and elucidating crosstalk between pathways. This short Review, with three figures and 70 citations, describes several representative examples of the application of these fluorescence-based tools and discusses future uses.
