Abstract

This Podcast features an interview with Amnon Altman, senior author of a Research Article that appears in the 30 September 2014 issue of Science Signaling, about how the guanine nucleotide exchange factor SLAT promotes calcium signaling in T cells. Binding of antigen to the T cell receptor leads to production of the second messenger IP3, which binds to IP3 receptor 1 (IP3R1) to stimulate calcium release from the endoplasmic reticulum. This initial release of calcium causes calcium channels at the plasma membrane to open, resulting in a sustained rise in intracellular calcium that triggers T cell activation. Fos et al. report that the guanine nucleotide exchange factor SLAT bound to IP3R1 to promote calcium release from the endoplasmic reticulum. Blocking the interaction between SLAT and IP3R1 with a peptide inhibited T cell activation. Drugs that block this interaction might be useful for reducing T cell activation in patients with diseases of autoimmunity or inflammation.
