The animation shows a schematic representation of how trans-endocytosis of Notch into Delta-expressing cells contributes to Notch activation. Notch proteins are receptors that mediate several developmental processes. Notch receptors are activated upon binding transmembrane ligands of the DSL family, such as Delta (δ). Following ligand binding, Notch is cleaved twice. First, Notch is cleaved in its extracellular domain at the S2 site. The subsequent cleavage within Notch's transmembrane domain at the S3 site releases an intracellular domain (N-ICD), which translocates into the nucleus and regulates the transcription of target genes. The cleavage and trans-endocytosis of the Notch extracellular domain (N-ECD) into the ligand-expressing cell facilitates release of the N-ICD.
